Estudos de infecções experimentais com alguns flavivírus demonstraram que esses possuem capacidade de causar persistência da infecção.  OBJETIVO: Investigar a infecção persistente pelo VILH em hamsters dourados (Mesocricetus auratus).  MATERIAIS E MÉTODOS: Os hamsters foram infectados por via intraperitoneal com VILH (9,6 DL50/0,02 mL), sendo, por um período de até 120 dias pós-inoculação (d.p.i.), anestesiados e sacrificados para coleta de amostras de sangue, urina e tecidos. Títulos virais para o VILH foram quantificados por transcrição reversa seguida de reação em cadeia mediada pela polimerase em tempo real (qRT-PCR) nas amostras biológicas, e as mesmas foram utilizadas para infectar células VERO, com confirmação da replicação viral por imunofluorescência. Níveis de anticorpos totais no soro foram determinados pela técnica inibição da hemaglutinação, as alterações teciduais observadas por meio do exame histopatológico corado por hematoxilina-eosina e detecção de antígenos virais por imuno-histoquímica.  RESULTADOS: O VILH induziu forte resposta imune, demonstrou ser patogênico para hamsters, causando invasão neuronal, foi isolado em células VERO de vísceras, cérebro, sangue, soro e urina dos hamsters infectados e detectado por qRT-PCR no cérebro, fígado e sangue com 30, seis e 15 d.p.i., respectivamente. As alterações histopatológicas e a expressão de antígenos virais, nas amostras de fígado, rim e pulmão, ocorreram com 30 d.p.i.; e no cérebro, até quatro meses pós-inoculação.  CONCLUSÃO: Os resultados obtidos mostraram que o VILH é patogênico para hamsters dourados e tem capacidade de causar infecção persistente em hamsters infectados por via periférica.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT  INTRODUCTION: Ilheus virus (ILHV) is a Brazilian arbovirus belonging to the Flaviviridae family and Flavivirus genus. The virus is clinically classified as encephalitogenic, and is known to cause infection persistence.  OBJECTIVE: To investigate persistent infection by ILHV in golden hamsters (Mesocricetus auratus).  MATERIALS AND METHODS: Hamsters were infected intraperitoneally with ILHV (9.6 LD50/0.02 mL), anaesthetised, and sacrificed up to the 120th day post-infection, for blood, urine, and tissue sampling. Viral titers were measured for ILHV using reverse transcription followed by real-time reverse transcription polymerase chain reaction (qRT-PCR) performed on biological samples, which were used to infect VERO cells; viral replication was confirmed by immunofluorescence. Serum antibody levels were determined using hemagglutination inhibition technique; tissue alterations were observed through histopathological examination of hematoxylin-eosin-stained tissues; viral antigens were detected using immunohistochemistry.  RESULTS: A strong immune response was induced by ILHV, in addition to its pathogenicity in hamsters and neural invasion. The virus was isolated from VERO cells in the viscera, brain, blood, serum, and urine of infected hamsters and detected by qRT-PCR in the brain, liver, and blood at 30, six, and 15 days post-infection, respectively. Histopathological alterations and detection of viral antigens in the liver, kidney, and lung samples occurred 30 days after infection; in the brain, they were detected up to four months after infection.  CONCLUSION: These results indicate that ILHV is pathogenic to golden hamsters and causes persistent infection in peripherally infected hamsters.]]></p></abstract>
<abstract abstract-type="short" xml:lang="es"><p><![CDATA[RESUMEN  INTRODUCCIÓN: El virus Ilhéus (VILH) es un arbovirus que ocurre en Brasil, perteneciente a la familia Flaviviridae, género Flavivirus, clasificado clínicamente como encefalitogénico. Estudios de infecciones experimentales con algunos flavivirus demostraron que tienen capacidad de causar la persistencia de la infección.  OBJETIVO: Investigar la infección persistente por el VILH en hámsteres dorados (Mesocricetus auratus).  MATERIAL Y MÉTODOS: Los hámsteres fueron infectados por vía intraperitoneal con VILH (9,6 DL50 / 0,02 mL), y por un período de hasta 120 días postinoculación (dpi) anestesiados y sacrificados para recolección de muestras de sangre, orina y tejidos. Títulos virales para el VILH fueron cuantificados por transcripción reversa seguida de reacción en cadena mediada por la polimerasa en tiempo real (qRT-PCR) en las muestras biológicas y las mismas se utilizaron para infectar células VERO, con confirmación de la replicación viral por inmunofluorescencia. Se determinaron los niveles de anticuerpos totales en suero con la técnica de inhibición de la hemoaglutinación, las alteraciones tisulares observadas en el examen histopatológico teñido por hematoxilina-eosina y la detección de antígenos virales por inmunohistoquímica.  RESULTADOS: el VILH indujo fuerte respuesta inmune, demostró ser patogénico para hámsteres, causando invasión neuronal, y fue aislado en células VERO de vísceras, cerebro, sangre, suero y orina de los hámsteres infectados y detectado por qRT-PCR en el cerebro, hígado y sangre con 30, 6 y 15 dpi., respectivamente. Las alteraciones histopatológicas y la expresión de antígenos virales en las muestras de hígado, riñón y pulmón sucedieron con 30 dpi.; y en el cerebro hasta cuatro meses postinoculación.  CONCLUSIÓN: Los resultados obtenidos mostraron que el VILH es patogénico para hámsteres dorados y tiene capacidad de causar infección persistente en hámsteres infectados por vía periférica.]]></p></abstract>
