Muitos microrganismos patogênicos e de vida livre que abrigam genes de resistência a antibióticos (ARGs) estão adaptados a habitats aquáticos, tornando os rios possíveis fontes de ARGs.  OBJETIVO:  descrever a ocorrência de ARGs segundo a variabilidade espacial e sazonal no lago da usina hidrelétrica Tucuruí.  MATERIAIS E MÉTODOS:  Amostras foram coletadas nas camadas fótica, afótica e sedimentar das estações Montante 1 e Montante Novo Repartimento, durante o verão e o inverno amazônicos. O DNA foi extraído e sequenciado na plataforma Ion Proton para análise metagenômica. A montagem de novo foi realizada com MEGAHIT; open reading frames foram preditas com Prodigal; e ARGs foram obtidos a partir das bases CARD e MEGARes. Contigs contendo ARGs foram gerados para análises posteriores. As análises estatísticas foram feitas no R, com o pacote ggplot2.  RESULTADOS:  Os ARGs mais abundantes no conjunto de amostras foram betalactâmicos (3.317,92 ×/Gb), macrolídeo-lincosamida-estreptogramina (1.079,77 ×/Gb), tetraciclina (968,23 ×/Gb), aminoglicosídeo (779,25 ×/Gb) e genes de resistência a múltiplas drogas (698,94 ×/Gb), com destaque de ocorrência sobretudo no inverno amazônico na camada superficial do lago.  CONCLUSÃO:  Os resultados indicam importante influência antrópica e preocupação com a disseminação de genes de resistência a antibióticos no lago da usina hidrelétrica Tucuruí.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT Tucuruí is the second largest fully Brazilian hydroelectric plant, and the artificial lake formed after its construction profoundly altered the regional ecosystem. Many pathogenic and free-living microorganisms that harbor antibiotic resistance genes (ARGs) are adapted to aquatic habitats, making rivers potential reservoirs of ARGs.  OBJECTIVE:  To describe the occurrence of ARGs according to spatial and seasonal variability in the Tucuruí Hydroelectric Plant reservoir.  MATERIALS AND METHODS:  Samples were collected from the photic, aphotic, and sediment layers at the Montante 1 and Montante Novo Repartimento stations during the Amazonian summer and winter. DNA was extracted and sequenced on the Ion Proton platform for metagenomic analysis. De novo assembly was performed using MEGAHIT, open reading frames were predicted with Prodigal, and ARGs were identified using the CARD and MEGARes databases. Contigs containing ARGs were generated for subsequent analyses. Statistical analyses were conducted in R using the ggplot2 package.  RESULTS:  The most abundant ARGs in the samples were those related to beta-lactams (3,317.92 ×/Gb), macrolide-lincosamide-streptogramin (1,079.77 ×/Gb), tetracyclines (968.23 ×/Gb), aminoglycosides (779.25 ×/Gb), and multidrug resistance genes (698.94 ×/Gb). These ARGs occurred predominantly during the Amazonian winter in the surface layer of the reservoir.  CONCLUSION:  The outcomes screen significant anthropogenic influence and concern regarding the spread of antibiotic resistance genes in the Tucuruí Hydroelectric Plant reservoir.]]></p></abstract>
