Com o objetivo de detectar e caracterizar cepas de HRV associadas a casos de síndrome respiratória aguda grave (SRAG) na Cidade de Belém, Estado do Pará, Brasil, foram analisadas 224 amostras de pacientes com SRAG atendidos em unidades hospitalares no período de janeiro de 2013 a janeiro de 2014. A análise das amostras foi desenvolvida utilizando-se três etapas principais: a) extração do RNA viral (RNAv); b) amplificação do RNAv pelas técnicas de reação em cadeia mediada pela polimerase precedida de transcrição reversa (RT-PCR) em tempo real e RT-PCR convencional; e c) sequenciamento do genoma viral. Entre as 224 amostras analisadas, 59 (26,3%) foram positivas para HRV; dessas, em 22 foi possível realizar a caracterização das espécies de HRV por sequenciamento, sendo 13 (59%) classificadas como HRV-A, oito (36,3%) como HRV-C; uma foi classificada como enterovírus humano D68 (EV-D68). A distribuição por faixa etária evidenciou que os adultos foram os mais acometidos pelo HRV, representando 45,7% (n = 27) dos casos positivos. Notou-se ainda a grande concentração de casos positivos para o vírus no grupo de 0-4 anos de idade durante o período estudado (n = 23, 39%). Quanto à distribuição mensal do HRV em Belém, foi verificada a circulação predominantemente no primeiro semestre do ano, a qual costuma estar associada ao período de maior pluviosidade na região. Os resultados obtidos expressam uma taxa de infecção por HRV relevante, mostrando que esse vírus é um agente importante no que diz respeito às infecções respiratórias em Belém.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT Human rhinovirus (HRV) is the most common viral agent associated with infections of the upper respiratory tract and is the main causative agent of the common cold. This study aimed to detect and characterize HRV strains associated with severe acute respiratory syndrome (SARS) in Belém, Pará State, Brazil, by analyzing samples from 224 SARS patients admitted to hospitals between January 2013 and January 2014. Sample analysis was performed in three stages: (a) viral RNA isolation (vRNA); (b) amplification of vRNA using conventional and real-time reverse transcription polymerase chain reaction; and (c) viral genome sequencing. Among the 224 analyzed samples, 59 (26.3%) were HRV-positive, of which 22 HRV species could be characterized through sequencing. Thirteen (59%) were classified as HRV-A and eight (36.3%) as HRV-C; one sample was classified as human enterovirus D68 (EV-D68). Distribution by age revealed that adults were the most affected by HRV, accounting for 45.7% (n = 27) of total positive cases. There was an increased number of HRV-positive cases in the 0-4-year-old group (n = 23, 39%) during the study period. Relating to monthly HRV distribution in Belém, circulation was predominant during the first semester, which is typically associated with increased rainfall. The results indicate a high HRV infection rate; thus indicating that the pathogen is an important agent of respiratory infections in Belém.]]></p></abstract>
<abstract abstract-type="short" xml:lang="es"><p><![CDATA[RESUMEN El Rinovirus humano (RVH) es el más común entre los agentes virales asociados a infecciones en el tracto respiratorio superior, siendo reconocido como el principal patógeno causador del resfriado común. Con el objetivo de detectar y caracterizar cepas de RVH asociadas a casos de síndrome respiratorio agudo grave (SRAG) en la ciudad de Belém, Pará, Brasil, se analizaron 224 muestras de pacientes con SRAG atendidos en unidades hospitalarias no período de enero de 2013 a enero de 2014. El análisis de las muestras se desarrolló utilizando tres etapas principales: a) extracción del ARN viral (ARNv); b) Amplificación del ARNv por las técnicas de RT-PCR en tiempo real y RT-PCR convencional; y c) secuenciación del genoma viral. Entre las 224 muestras analizadas 59 (26,3%) fueron positivas para RVH, de estas, en 22 fue posible realizar la caracterización de las especies de RVH por secuenciación. Siendo 13 (59%) clasificadas como RVH-A y ocho (36,3%) como RVH-C. La distribución por grupo de edad pone en evidencia que los adultos fueron los más acometidos por el RVH expresando 45,7% (n=27) de los casos positivos. Se nota además la gran concentración de casos positivos para el virus en el grupo de cero a cuatro años durante el período estudiado (n=23, 39%). Con relación a la distribución mensual del RVH en la ciudad de Belém, se comprobó su circulación, predominantemente, en el primer semestre del año, lo que suele estar asociado al período de mayor pluviosidad en nuestra Región. Los resultados obtenidos expresan una tasa de infección por RVH relevante, mostrando que ese virus es un importante agente en lo que dice respecto a las infecciones respiratorias en la ciudad de Belém, Pará, Brasil.]]></p></abstract>
