A maioria dos arbovírus está classificada na família Bunyaviridae que, por possuírem três segmentos de RNA, apresentam uma característica importante de rearranjo genético, formando novos vírus no mundo. O advento de novas ferramentas de biologia molecular para sequenciamento genômico, aliado aos clássicos testes sorológicos de fixação do complemento (FC), teste de neutralização (TN) e inibição da hemaglutinação (IH) permitem uma melhor caracterização e identificação viral, principalmente dos vírus frutos de rearranjos genéticos. Dessa forma, este estudo descreve as características sorológicas e moleculares de três isolados virais (BeAR 544767, BeAR 643361 e BeAR 701402) obtidos a partir de mosquitos no Estado do Pará pela Seção de Arbovirologia e Febres Hemorrágicas do Instituto Evandro Chagas. Os testes de FC e TN foram utilizados para a caracterização antigênica e, para a molecular, foram utilizadas duas plataformas de sequenciamento genômico: 454 FLX e Ion PGMTM. Pelo teste de FC foi possível definir que os três isolados apresentavam reação cruzada entre os vírus Tucunduba (VTUC) e vírus Taiassui. O TN definiu os isolados virais BeAR 544767 e BeAR 701402 como cepas do VTUC, sendo essa classificação confirmada também pela caracterização molecular para os três isolados virais em estudo. Portanto, os isolados virais BeAR 544767, BeAR 643361 e BeAR 701402 foram classificados como cepas do VTUC, dentro do grupo Wyeomyia, família Bunyaviridae, gênero Orthobunyavirus.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT Arboviruses are widely distributed worldwide and, in Brazil, approximately 200 species have been isolated and many are associated with human diseases. Most arboviruses are classified within the Bunyaviridae family, which present three RNA segments and an important genetic reassortment characteristic, thus forming new types of viruses globally. New molecular biology techniques for genome sequencing, associated with classic serological tests, such as complement fixation (CF), neutralization test (NT), and hemagglutination inhibition (HI), enable enhanced viral identification and characterization, particularly of viruses generated by genetic reassortment. Thus, the serological and molecular characteristics of three viral samples (BeAR 544767, BeAR 643361, and BeAR 701402) obtained from mosquitoes in Pará State by the Arbovirology and Hemorrhagic Fevers Section of the Instituto Evandro Chagas were determined. Antigen characterization was performed using CF and NT; molecular characterization was conducted using two genomic sequencing platforms: 454 FLX and Ion PGMTM. The results of CF indicated that all three isolates presented cross-reactivity between the Tucunduba (TUCV) and Taiassui viruses. Viral isolates of BeAR 544767 and BeAR 701402 were classified as TUCV strains by NT, which was confirmed by molecular characterization of the three isolates. Thus, viral isolates of BeAR 544767, BeAR 643361, and BeAR 701402 were classified as TUCV strains and they belong to the Wyeomyia group, Bunyaviridae family, and Orthobunyavirus genus.]]></p></abstract>

La mayoría de los arbovirus está clasificada en la familia Bunyaviridae, que por tener tres segmentos de ARN, presentan una característica importante de reacomodación genética, formando nuevos virus en todo el mundo. El surgimiento de nuevas herramientas de biología molecular para secuenciación genómica aliado a las clásicas pruebas serológicas de fijación del complemento (FC), neutralización (TN) e inhibición de la hemoaglutinación (HI), permiten una mejor caracterización e identificación viral, principalmente de los virus frutos de reacomodación genética. Así, este estudio describe las características serológicas y moleculares de tres aislados virales (BeAR 544767, BeAR 643361 y BeAR 701402) obtenidos a partir de mosquitos en el Estado de Pará por la Sección de Arbovirología y Fiebres Hemorrágicas del Instituto Evandro Chagas.
